Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan II is a synthetic peptide analog modeled on alpha-melanocyte-stimulating hormone, a naturally occurring signaling peptide involved in pigmentation. Its structure is a cyclic heptapeptide containing two non-natural substitutions, norleucine at position four and D-phenylalanine at position seven. These modifications resist enzymatic breakdown and extend the molecule's activity relative to the native hormone. The compound binds melanocortin receptors and is studied mainly as a pharmacological tool rather than a therapeutic product. It has never received approval as a medicine in any major jurisdiction.
The compound was developed in the late 1980s and 1990s by academic researchers investigating photoprotection. The rationale held that stimulating melanin production might reduce ultraviolet damage to skin and lower skin cancer risk. Early work examined receptor binding, pigment response, and short-term tolerability in small studies. That program did not produce an approved drug, and formal development stalled after early-phase trials. Whether induced pigmentation confers meaningful photoprotection remains an open question.
On December 24, 1851, the largest fire in the library's history destroyed 35,000 books, two-thirds of the library's collection, and two-thirds of Thomas Jefferson's original transfer. Congress appropriated $168,700 to replace the lost books in 1852 but not to acquire new materials. (By 2008, the librarians of Congress had found replacements for all but 300 of the works that had been documented as being in Jefferson's original collection.) This marked the start of a conservative period in the library's administration by librarian John Silva Meehan and joint committee chairman James A. Pearce, who restricted the library's activities. Meehan and Pearce's views about a restricted scope for the Library of Congress reflected those shared by members of Congress. While Meehan was a librarian, he supported and perpetuated the notion that "the congressional library should play a limited role on the national scene and that its collections, by and large, should emphasize American materials of obvious use to the U.S. Congress." In 1859, Congress transferred the library's public document distribution activities to the Department of the Interior and its international book exchange program to the Department of State. During the 1850s, Smithsonian Institution librarian Charles Coffin Jewett aggressively tried to develop the Smithsonian as the United States national library. His efforts were rejected by Smithsonian secretary Joseph Henry, who advocated a focus on scientific research and publication.
=== Blood tests === When RA is clinically suspected, a physician may test for rheumatoid factor (RF) and anti-citrullinated protein antibodies (ACPAs measured as anti-CCP antibodies). The test is positive approximately two-thirds of the time, but a negative RF or CCP antibody does not rule out RA; rather, the arthritis is called seronegative, which occurs in approximately a third of people with RA. During the first year of illness, rheumatoid factor is more likely to be negative with some individuals becoming seropositive over time. RF is a non-specific antibody and seen in about 10% of healthy people, in many other chronic infections like hepatitis C, and chronic autoimmune diseases such as Sjögren's disease and systemic lupus erythematosus. Therefore, the test is not specific for RA. Hence, new serological tests check for anti-citrullinated protein antibodies ACPAs. These tests are again positive in 61–75% of all RA cases, but with a specificity of around 95%. As with RF, ACPAs are many times present before symptoms have started. The by far most common clinical test for ACPAs is the anti-cyclic citrullinated peptide (anti CCP) ELISA. In 2008, a serological point-of-care test for the early detection of RA combined the detection of RF and anti-MCV with a sensitivity of 72% and specificity of 99.7%. To improve the diagnostic capture rate in the early detection of patients with RA and to risk-stratify these individuals, the rheumatology field continues to seek complementary markers to both RF and anti-CCP.
=== Acute toxicity === Melamine is reported to have an oral median lethal dose (LD50) of 3248 mg/kg based on rat data. It is also an irritant when inhaled or in contact with the skin or eyes. The reported dermal LD50 is greater than 1000 mg/kg for rabbits. A study by Soviet researchers in the 1980s suggested that melamine cyanurate, commonly used as a fire retardant, could be more toxic than either melamine or cyanuric acid alone. For rats and mice, the reported LD50 for melamine cyanurate was 4.1 g/kg (given inside the stomach) and 3.5 g/kg (via inhalation), compared to 6.0 and 4.3 g/kg for melamine and 7.7 and 3.4 g/kg for cyanuric acid respectively. A toxicology study in animals conducted after recalls of contaminated pet food concluded that the combination of melamine and cyanuric acid in diet does lead to acute kidney injury in cats. A 2008 study produced similar experimental results in rats and characterized the melamine and cyanuric acid in contaminated pet food from the 2007 outbreak. A 2010 study from Lanzhou University attributed kidney failure in humans to uric acid stone accumulation after ingestion of melamine resulting in a rapid aggregation of metabolites such as cyanuric acid diamide (ammeline) and cyanuric acid. A 2013 study demonstrated that melamine can be metabolized to cyanuric acid by gut bacteria. In particular, Klebsiella terrigena was determined to be a factor in melamine toxicity. In culture, K. terrigena was shown to convert melamine to cyanuric acid directly.
== Corneal epithelial cells == Corneal epithelial cells present a physical barrier to prevent microbes from reaching the interior of the eye chamber, which is effectively separated from the rest of the body via tight junctions. At the same time, corneal epithelial cells also secrete cytokines to activate microbial defense. One cytokine, interleukin (IL)-1α, is stored in epithelial cells and automatically released when the cell membrane is ruptured by infection or trauma. However, long-term effects of IL-1α can lead to not only enhanced immune infiltration of the cornea, but also neovascularization (formation of new blood vessels), which can lead to a loss of corneal transparency. Therefore, the cornea has also been found to secrete an IL-1α antagonist, IL-1RN, which decreases leucocyte invasion of the cornea and suppresses neovascularization, both of which can help preserve vision.

== Biosynthesis == Gramicidin S biosynthetic pathway consists of two-enzyme of nonribosomal peptide synthases (NRPSs), gramicidin S synthetase I (GrsA) and gramicidin S synthetase II (GrsB), to give a product as a cyclic decapeptide. Within the biosynthetic pathway, there are total of five modules that specifically recognize, activate, and condense the amino acids to gramicidin S. Starting module GrsA consists of three domains: Adenylation (A) domain where it incorporates the amino acid and activates it by adenylation using ATP, Thiolation (T) domain or peptidyl carrier protein (PCP) in which the adenylated amino acid gets covalently attached to the 4´-phosphopantetheine group and this gets loaded onto the conserved serine in the T domain, Epimerization (E) domain where it epimerizes L-amino acid to D-amino acid. Starting module GrsA loads D-Phe onto the system. Second enzyme cluster GrsB contains four modules, each containing condensation (C), adenylation (A), and thiolation (T) domains and thioesterase domain (TE) at the end. C domain forms a peptide bond between two amino acids, D-Phe and L-Pro. L-Val, L-Orn, and L-Leu are incorporated sequentially by the next three modules of GrsB. After repeating the whole module synthesis once again, TE domain cyclizes and releases the two peptides and dimerize them together to form the final product.
On average, male mammals are larger than females, with males being at least 10% larger than females in over 45% of investigated species. Most mammalian orders also exhibit male-biased sexual dimorphism, although some orders do not show any bias or are significantly female-biased (Lagomorpha). Sexual size dimorphism increases with body size across mammals (Rensch's rule), suggesting that there are parallel selection pressures on both male and female size. Male-biased dimorphism relates to sexual selection on males through male–male competition for females, as there is a positive correlation between the degree of sexual selection, as indicated by mating systems, and the degree of male-biased size dimorphism. The degree of sexual selection is also positively correlated with male and female size across mammals. Further, parallel selection pressure on female mass is identified in that age at weaning is significantly higher in more polygynous species, even when correcting for body mass. Also, the reproductive rate is lower for larger females, indicating that fecundity selection selects for smaller females in mammals. Although these patterns hold across mammals as a whole, there is considerable variation across orders.
Robert William Barker (December 12, 1923 – August 26, 2023) was an American media personality, game show host, and animal rights advocate. He hosted CBS's The Price Is Right, the longest-running game show in North American television history, from 1972 to 2007. Barker also hosted Truth or Consequences from 1956 to 1975. Born in Darrington, Washington, in modest circumstances, Barker spent most of his youth on the Rosebud Indian Reservation and was a citizen of the Rosebud Sioux Tribe. Barker joined the United States Navy Reserve during World War II. He worked part-time in radio while attending college. In 1950, Barker moved to California to pursue a broadcasting career. He was given his own radio show, The Bob Barker Show, which ran for six years. Barker began his game show career in 1956, hosting Truth or Consequences. Barker began hosting The Price Is Right in 1972. He became an advocate for animal rights and of animal rights activism, supporting groups such as the United Activists for Animal Rights, People for the Ethical Treatment of Animals, and the Sea Shepherd Conservation Society. In 2007, Barker retired from hosting The Price Is Right after celebrating his 50-year career on television. Regarded as a pop culture icon, Barker continued to make occasional appearances for several years into his retirement until 2015.
== Production == Like many aspects of nuclear power, fast breeder reactors have been subject to much controversy over the years. In 2010 the International Panel on Fissile Materials said "After six decades and the expenditure of the equivalent of tens of billions of dollars, the promise of breeder reactors remains largely unfulfilled and efforts to commercialize them have been steadily cut back in most countries". In Germany, the United Kingdom, and the United States, breeder reactor development programs have been abandoned. The rationale for pursuing breeder reactors—sometimes explicit and sometimes implicit—was based on the following key assumptions:
== Contraindications == Hypersensitivity to dalbavancin can occur, causing issues such as skin reactions or anaphylaxis. Caution is advised for patients with known hypersensitivity to other glycopeptides. There is currently no data on cross-reactivity between dalbavancin and vancomycin.

=== Mass campaigns and high office === Liu retained his political standing through active participation in the state's major mass campaigns. During the land reform movement, Liu announced at a conference that: "I once used to be a big landlord, but now I will unconditionally and unreservedly distribute the land to the peasants." According to Guangming Daily, this statement won him enthusiastic applause from the attendance. In 1955, during the Sufan campaign, Liu called for "greater vigilance" against "counterrevolutionaries". In 1956, he spoke in Chengdu on the occasion of the 90th anniversary of Sun Yat-sen's birth, publicly praising all of the Communist Party's political positions. Politically, he was elected to the 3rd Central Committee of the Revolutionary Committee of the Chinese Kuomintang in 1954, serving until his death. He also served as Minister of Forestry from 1959 to 1967. In this capacity, he made reports on the state of forestry in China and carried out inspections in Jiangxi province as well as other regions. At the second National People's Congress in 1960 during the Great Leap Forward, Liu Wenhui declared that the country was in the midst of a "forestry revolution." Liu Wenhui escaped persecution during the Cultural Revolution due to Zhou Enlai's protection, but he was unable to stop his older brother, Liu Wencai (who had died in 1949), from becoming demonized by state media and remembered in the popular imagination as a tyrannical and corrupt landlord.
===== Morphology change ===== As shown by flow cytometry and electron microscopy, the most sensitive sign of activation, when exposed to platelets using ADP, are morphological changes. Mitochondrial hyperpolarization is a key event in initiating morphology changes. Intraplatelet calcium concentration increases, stimulating the interplay between the microtubule/actin filament complex. The continuous changes in shape from the unactivated to the fully activated platelet are best seen via scanning electron microscopy. The three steps along this path are named early dendritic, early spread, and spread. The surface of the unactivated platelet looks similar to the surface of the brain–a wrinkled appearance from numerous shallow folds that increase the surface area; early dendritic, an octopus with multiple arms and legs; early spread, an uncooked frying egg in a pan, the "yolk" is the central body; and the spread, a cooked fried egg with a denser central body. These changes are all brought about by the interaction of the microtubule/actin complex with the platelet cell membrane and open canalicular system (OCS), which is an extension and invagination of that membrane. This complex runs just beneath these membranes and is the chemical motor that pulls the invaginated OCS out of the interior of the platelet, like turning pants pockets inside out, creating the dendrites. This process is similar to the mechanism of contraction in a muscle cell. The entire OCS thus becomes indistinguishable from the initial platelet membrane as it forms the "fried egg".
=== Ethics === When considering any of these methods of treatment ethical concerns should also be factored into the assessment. Textiles may have historical and cultural significance that conflict with their status as a museum object. In these cases, it is important to connect with the appropriate experts to create a plan that respects any cultural heritage boundaries while honoring a commitment to preserve and display objects in alignment with an institution's mission statement.
Live Science reported that lead author of the study Matteo Borrini from Liverpool John Moores University stated: "these cannot be real bloodstains from a person who was crucified and then put into a grave, but actually handmade by the artist that created the shroud." In 2025 a study was published in the journal Archaeometry, by Brazilian digital graphics expert and 3D designer Cicero Moraes. Moraes used software to model how clothing would move on a three-dimensional human body compared to a low-relief sculpture. In Moraes’s experiments, the image produced when a cloth is draped over a 3D human model appears misshapen and distorted. This is called the Agamemnon Mask effect, named after the Mycenaean gold funerary mask. Moraes also found that the imprint made from a low-relief sculpture closely matched the image on the Turin shroud. This supports his conclusion that the image on the shroud was an artistic creation, which Moraes interpreted as being a funerary object and a "masterwork of Christian art."

No. It is a laboratory-made peptide, while the natural hormone is alpha-melanocyte-stimulating hormone, a longer peptide produced by the pituitary gland and by skin cells. Melanotan-2 mimics only a short active region of that hormone and contains non-natural residues such as D-phenylalanine.
No approved regulatory indication exists in major markets. An approval exists for a different peptide, afamelanotide, which is used for a rare photosensitivity disorder called erythropoietic protoporphyria. Melanotan-2 itself remains a research compound with no cleared clinical role.
Melanocortin-1 receptors on melanocytes are the receptor most closely linked to pigment production. The peptide is not selective, however, and also activates melanocortin-3, melanocortin-4, and melanocortin-5 receptors. That lack of selectivity is the usual explanation offered for both its range of observed effects and its off-target effects.
It is a synthetic cyclic heptapeptide and a non-selective melanocortin receptor agonist. Structurally it is modelled on alpha-melanocyte-stimulating hormone, a peptide that occurs naturally in the body.
