The short version of synthetic peptide fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-08-01. Anything still debated is marked as such rather than presented as settled.
The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.

=== Activated charcoal adsorption === Granular activated carbon filtering utilizes a form of activated carbon with a high surface area, and adsorbs many compounds, including many toxic compounds. Water passing through activated carbon is commonly used in concert with hand pumped filters to address organic contamination, taste, or objectionable odors. Activated carbon filters are not usually used as the primary purification techniques of portable water purification devices, but rather as secondary means to complement another purification technique. It is most commonly implemented for pre- or post-filtering, in a separate step than ceramic filtering, in either case being implemented prior to the addition of chemical disinfectants used to control bacteria or viruses that filters cannot remove. Activated charcoal can remove chlorine from treated water, removing any residual protection remaining in the water protecting against pathogens, and should not, in general, be used without careful thought after chemical disinfection treatments in portable water purification processing. Ceramic/Carbon Core filters with a 0.5 μm or smaller pore size are excellent for removing bacteria and cysts while also removing chemicals.
=== Heart rate and energy expenditure === Studies have shown that compared to wild type mice, Ffar3 gene knockout mice have: a) significantly smaller-sized sympathetic nervous system ganglia as judged by measurements of this systems' largest ganglia, the superior cervical ganglion; b) significantly slower heart rates; and c) significantly lower norepinephrine levels in their blood plasma. (Norepinephrine is a neurotransmitter that is released by sympathetic nervous system neurons and among other actions increases heart rate and total body energy expenditure.) Furthermore, the treatment of wild type mice with propionic acid increased their heart rates but did not do so in Ffar3 gene knockout mice. Finally, the offspring of Ffar3-gene knockout mice had slower heart rates (as well as lower body temperatures) than the offspring of wild type mice. These findings indicate that FFAR3 regulates heart rates and energy expenditure in mice. Studies are needed to determine if it does so in humans.
Recent advancements in bioorthogonal chemistry have revealed applications in protein analysis. The extension of using organic molecules to observe their reaction with proteins reveals extensive methods to tag them. Unnatural amino acids and various functional groups represent new growing technologies in proteomics. Specific biomolecules that are capable of being metabolized in cells or tissues are inserted into proteins or glycans. The molecule will have an affinity tag, modifying the protein allowing it to be detected. Azidohomoalanine (AHA) utilizes this affinity tag via incorporation with Met-t-RNA synthetase to incorporate into proteins. This has allowed AHA to assist in determine the identity of newly synthesized proteins created in response to perturbations and to identify proteins secreted by cells. Recent studies using ketones and aldehydes condensations show that they are best suited for in vitro or cell surface labeling. However, using ketones and aldehydes as bioorthogonal reporters revealed slow kinetics indicating that while effective for labeling, the concentration must be high. Certain proteins can be detected via their reactivity to azide groups. Non-proteinogenic amino acids can bear azide groups which react with phosphines in Staudinger ligations. This reaction has already been used to label other biomolecules in living cells and animals. The bioorthogonal field is expanding and is driving further applications within proteomics. It is worthwhile noting the limitations and benefits.
In the 1960s, Australia - particularly Queensland - launched the first awareness campaign on the dangers of ultraviolet (UV) radiation in the spirit of primary prevention. In the 1980s, many countries in Europe and overseas initiated similar UV protection campaigns. UV radiation has a thermal effect on the skin and eyes and can lead to skin cancer (malignant melanoma) and eye inflammation or cataracts. To protect the skin from harmful UV radiation, such as photodermatosis, acne aestivalis, actinic keratosis or urticaria solaris, normal clothing, special UV protective clothing (SPF 40-50) and high SPF sunscreen can be used. The Australian-New Zealand Standard (AS/NZS 4399) of 1996 measures new textile materials in an unstretched and dry state for the manufacture of protective clothing worn while bathing, especially by children, and for the manufacture of shading textiles (sunshades, awnings). The UV Standard 801 assumes a maximum radiation intensity with the solar spectrum in Melbourne, Australia, on January 1 of a year (at the height of the Australian summer), the most sensitive skin type of the wearer, and under wearing conditions. As the solar spectrum in the northern hemisphere differs from that in Australia, the measurement method according to the European standard EN 13758-1 is based on the solar spectrum of Albuquerque (New Mexico, USA), which corresponds approximately to that of southern Europe. To protect your eyes, wear sunglasses with UV protection or special goggles that also shield the sides to prevent snow blindness.

=== Illegal drug trafficking === Drug trafficking sales have gone up in Colombia since the 1970s due to the conflict. Many Mexican drug cartels have been spotted operating in the area, including the Sinaloa Cartel, Gulf Cartel, Beltrán Leyva Cartel, CJNG, and Los Zetas. The Medellín Cartel and Cali Cartel were the first drug trafficking organizations in Colombia and had alliances with the Guadalajara Cartel, Los Valencia Organization, and Amezcua Cartel.
== History == Mid-1990s: Oracle Clinical is introduced as part of Oracle’s life-sciences applications portfolio (first documentation references appear in 1996). Adoption in large sponsors (late 1990s–2000s): Deployment at major companies; for example, Boehringer Ingelheim reported selecting Oracle Clinical/RDC as its EDC to integrate with existing clinical databases and scale to most trials. RDC Classic → RDC Onsite (2000s): A web-based interface (RDC Onsite) supersedes the legacy client, enabling browser-based site entry and monitoring documented by sponsors and public research networks. Acquisition era (2009–2012): Oracle expands the health-sciences portfolio with acquisitions of Relsys (Argus Safety, 2009), Phase Forward (2010), and ClearTrial (2012), shaping the broader eClinical suite around OC/RDC. Cloud transition (2020s): Oracle introduces Clinical One Data Collection as a cloud-native path for trial data capture, positioned alongside or as a successor for on-premises OC/RDC deployments.
=== Cleavage and ligation === In addition to those listed above, the most important modification of primary structure is peptide cleavage (by chemical hydrolysis or by proteases). Proteins are often synthesized in an inactive precursor form; typically, an N-terminal or C-terminal segment blocks the active site of the protein, inhibiting its function. The protein is activated by cleaving off the inhibitory peptide. Some proteins even have the power to cleave themselves. Typically, the hydroxyl group of a serine (rarely, threonine) or the thiol group of a cysteine residue will attack the carbonyl carbon of the preceding peptide bond, forming a tetrahedrally bonded intermediate [classified as a hydroxyoxazolidine (Ser/Thr) or hydroxythiazolidine (Cys) intermediate]. This intermediate tends to revert to the amide form, expelling the attacking group, since the amide form is usually favored by free energy, (presumably due to the strong resonance stabilization of the peptide group). However, additional molecular interactions may render the amide form less stable; the amino group is expelled instead, resulting in an ester (Ser/Thr) or thioester (Cys) bond in place of the peptide bond. This chemical reaction is called an N-O acyl shift. The ester/thioester bond can be resolved in several ways:
=== Medical === As of April 2019, the US Food and Drug Administration (FDA) had stated that there were no approved clinical uses for kratom, and that there was no evidence that kratom was safe or effective for treating any condition. This reiterated the conclusion of an earlier report by the European Monitoring Centre for Drugs and Drug Addiction (EMCDDA): As of 2023, mitragynine had not been approved for any medical use. As of 2018, the FDA had noted, in particular, that there had been no clinical trials to study safety and efficacy of kratom in the treatment of opioid addiction.

=== Black box warning === In January 2012, the FDA announced that because brentuximab vedotin had been linked with two cases of progressive multifocal leukoencephalopathy, they were requiring the addition of a black box warning to the drug label regarding this potential risk.
The Nirenberg and Matthaei experiment was a scientific experiment performed in May 1961 by Marshall W. Nirenberg and his post-doctoral fellow, J. Heinrich Matthaei, at the National Institutes of Health (NIH). The experiment deciphered the first of the 64 triplet codons in the genetic code by using nucleic acid homopolymers to translate specific amino acids. In the experiment, an extract was prepared from bacterial cells that could make protein without the presence of intact living cells. An artificial form of RNA consisting entirely of uracil-containing nucleotides (polyuridylic acid or poly-U) was added to the extract, causing it to form a protein composed entirely of the amino acid phenylalanine. This experiment cracked the first codon of the genetic code and showed that RNA controlled the production of specific types of protein.
However, Georgiev already has limited influence in the government and pressure against Velchev was increasing. On 2 July the Communists passed for one day a law on control of the army, seizing powers from the war minister at the expense of the Council of Ministers. On 2 August, Kimon Georgiev agreed to the removal of his close associates Kiril Stanchev (arrested and convicted in a show trial) and Damyan Velchev (effectively replaced as minister by Krum Lekarski and interned). From 11 August to 3 September, Kimon Georgiev was in Paris at the head of a Bulgarian delegation preparing the Paris Peace Treaty. There he met with various politicians, including the foreign ministers of the Soviet Union, Vyacheslav Molotov, and the United States, James Byrnes. Bulgaria unsuccessfully insisted on being recognized as a country that had fought against Germany, but with the support of the Eastern Bloc countries, it managed to significantly reduce the reparations demanded by Greece. Upon his return to Bulgaria, Kimon Georgiev managed to personally solicit protections for Damyan Velchev from Georgi Dimitrov. On 25 September, Georgiev nominally became head of the War Ministry, and Velchev was sent as ambassador to Switzerland, avoiding for the moment a show trial. In September–November 1946, Georgiev was in charge of the War Ministry. During this period, he was promoted to the rank of colonel general. On 5 October 1946, at a rally in Sofia, Kimon Georgiev opened the Fatherland Front's election campaign for the election of the Sixth Grand National Assembly.
== Prevention == Many types of dwarfism are currently impossible to prevent because they are genetically caused. Genetic conditions that cause dwarfism may be identified with genetic testing, by screening for the specific variations that result in the condition. However, due to the number of causes of dwarfism, it may be impossible to determine definitively if a child will be born with dwarfism. Dwarfism resulting from malnutrition or a hormonal abnormality may be treated with an appropriate diet or hormonal therapy. Growth hormone deficiency may be remedied via injections of human growth hormone during childhood.
==== Phycoerythrin-like proteins ==== Phycobiliproteins are water soluble light-capturing proteins, produced by cyanobacteria, and several algae. These pigments have been explored as fluorescent tags, food coloring agents, cosmetics, and immunological diagnostic agents. Most of these pigments are synthesized and accumulated intracellularly. As an exception, the cyanobacteria Oscillatoria and Scytonema sp. release an extracellular phycoerythrin-like 250 kDa protein. This pigment inhibits the growth of the green algae Chlorella fusca and Chlamydomonas and can be potentially used as an algicide.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
