Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Scientific discussion of Melanotan-2 spans pharmacology, dermatology, and public-health literature. Laboratory studies examine its receptor binding and cellular effects, while clinical reports describe outcomes observed after unregulated use. These two bodies of work differ in rigour and intent. Peer-reviewed trials of the compound as a medicine are limited, so much of the available information comes from case reports and surveillance data. Authors frequently note the gap between experimental findings and real-world use.
Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.

Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.
=== Nitric oxide === Nitric oxide (NO) is a major source of immunomodulation in rodents, and is produced by enzyme nitric oxide synthetase type 2 (NOS2) in the alveolar macrophage. NO inhibits tyrosine phosphorylation of the kinases involved in production of the interleukin-2 (IL-2) receptor, the expression of which is fundamental for T cell proliferation. In humans, however, NOS2 activity has been difficult to verify. There are two explanations for the lack of responsiveness in the promoter of human inducible nitric oxide synthetase (iNOS) to NO activation by lipopolysaccharides (LPS) + interferon gamma (IFNγ). The first is that there are various inactivating nucleotide variations in the human counterpart of the enhancer element that regulates LPS/IFNγ induced expression of the mouse NOS2 gene. The second is because of the absence of a nuclear factor in human macrophages that is required for optimum expression of gene NOS2 (LPS-inducible nuclear factor-kappa B/Rel complex). It is assumed that the difficulty in verifying NOS2 is due to a much more tightly controlled expression in human AMs as compared to that in the rodent AMs. NOS2 is part of an autoregulatory feedback loop, wherein an allergen or provoker stimulates inflammatory cytokine production, which in turn stimulates NO production, and NO down-regulates cytokine production.
Trifluridine (also called trifluorothymidine; abbreviation TFT or FTD) is an anti-herpesvirus antiviral drug, used primarily as prescription eyedrops. It was sold under the trade name Viroptic by Glaxo Wellcome, now merged into GlaxoSmithKline. The brand is now wholly owned by King Pharmaceuticals. Trifluridine was approved for medical use in 1980. It is also a component of the anti-cancer drug trifluridine/tipiracil, which is taken by mouth.
In turn, this leads to warmer sea surface temperatures (called El Niño), a weaker Walker circulation (an east—west overturning circulation in the atmosphere) and even weaker trade winds. Ultimately, the warm waters in the western tropical Pacific are depleted enough so that conditions return to normal. The exact mechanisms that cause the oscillation are unclear and are being studied. Each country that monitors the ENSO has a different threshold for what constitutes an El Niño or La Niña event, which is tailored to their specific interests. El Niño and La Niña affect the global climate and disrupt normal weather patterns, which can lead to intense storms in some places and droughts in others. El Niño events cause short-term (approximately one year in length) spikes in global average surface temperature, while La Niña events cause short term surface cooling. Therefore, the relative frequency of El Niño compared to La Niña events can affect global temperature trends on timescales of around ten years. The countries most affected by ENSO are developing countries that are bordering the Pacific Ocean and dependent on agriculture and fishing. In climate change science, ENSO is known as one of the internal climate variability phenomena. Future trends in ENSO due to climate change are uncertain, although climate change exacerbates the effects of droughts and floods.
The closure temperature or blocking temperature represents the temperature below which the mineral is a closed system for the studied isotopes. If a material that selectively rejects the daughter nuclide is heated above this temperature, any daughter nuclides that have been accumulated over time will be lost through diffusion, resetting the isotopic "clock" to zero. As the mineral cools, the crystal structure begins to form and diffusion of isotopes is less easy. At a certain temperature, the crystal structure has formed sufficiently to prevent diffusion of isotopes. Thus an igneous or metamorphic rock or melt, which is slowly cooling, does not begin to exhibit measurable radioactive decay until it cools below the closure temperature. The age that can be calculated by radiometric dating is thus the time at which the rock or mineral cooled to closure temperature. This temperature varies for every mineral and isotopic system, so a system can be closed for one mineral but open for another. Dating of different minerals and/or isotope systems (with differing closure temperatures) within the same rock can therefore enable the tracking of the thermal history of the rock in question with time, and thus the history of metamorphic events may become known in detail. These temperatures are experimentally determined in the lab by artificially resetting sample minerals using a high-temperature furnace. This field is known as thermochronology or thermochronometry.

On April 4, 2011, Texas Instruments announced that it had agreed to buy National Semiconductor for $6.5 billion in cash. Texas Instruments paid $25 per share of National Semiconductor stock, an 80% premium over the April 4, 2011, closing share price of $14.07. The deal made Texas Instruments one of the world's largest makers of analog technology components. On September 19, 2011, the Chinese minister approved the merger, the last one needed. The companies formally merged on September 23, 2011.
== Causes == Adverse drug reactions are classified as type A (intrinsic or pharmacological) or type B (idiosyncratic). Type A drug reaction accounts for 80% of all toxicities. Drugs or toxins that have a pharmacological (type A) hepatotoxicity are those that have predictable dose-response curves (higher concentrations cause more liver damage) and well characterized mechanisms of toxicity, such as directly damaging liver tissue or blocking a metabolic process. As in the case of paracetamol overdose, this type of injury occurs shortly after some threshold for toxicity is reached. Carbon tetrachloride is commonly used to induce acute type A liver injury in animal models. Idiosyncratic (type B) injury occurs without warning, when agents cause non-predictable hepatotoxicity in susceptible individuals, which is not related to dose and has a variable latency period. This type of injury does not have a clear dose-response nor temporal relationship, and most often does not have predictive models. Idiosyncratic hepatotoxicity has led to the withdrawal of several drugs from market even after rigorous clinical testing as part of the FDA approval process; Troglitazone (Rezulin) and trovafloxacin (Trovan) are two prime examples of idiosyncratic hepatotoxins pulled from market. The herb kava has caused a number of cases of idiosyncratic liver injury, ranging everywhere from asymptomatic to fatal.
=== Evolution === Genomic tRNA content is a differentiating feature of genomes among biological domains of life: Archaea present the simplest situation in terms of genomic tRNA content with a uniform number of gene copies, Bacteria have an intermediate situation and Eukarya present the most complex situation. Eukarya present not only more tRNA gene content than the other two kingdoms but also a high variation in gene copy number among different isoacceptors, and this complexity seem to be due to duplications of tRNA genes and changes in anticodon specificity. Evolution of the tRNA gene copy number across different species has been linked to the appearance of specific tRNA modification enzymes (uridine methyltransferases in Bacteria, and adenosine deaminases in Eukarya), which increase the decoding capacity of a given tRNA. As an example, tRNAAla encodes four different tRNA isoacceptors (AGC, UGC, GGC and CGC). In Eukarya, AGC isoacceptors are extremely enriched in gene copy number in comparison to the rest of isoacceptors, and this has been correlated with its A-to-I modification of its wobble base. This same trend has been shown for most amino acids of eukaryal species. Indeed, the effect of these two tRNA modifications is also seen in codon usage bias. Highly expressed genes seem to be enriched in codons that are exclusively using codons that will be decoded by these modified tRNAs, which suggests a possible role of these codons—and consequently of these tRNA modifications—in translation efficiency. Many species have lost specific tRNAs during evolution.
The two DNA strands then become templates for DNA polymerase to enzymatically assemble a new DNA strand from free nucleotides, the building blocks of DNA. As PCR progresses, the DNA generated is itself used as a template for replication, setting in motion a chain reaction in which the original DNA template is exponentially amplified. Almost all PCR applications employ a heat-stable DNA polymerase, such as Taq polymerase, an enzyme originally isolated from the thermophilic bacterium Thermus aquaticus. If the polymerase used was heat-susceptible, it would denature under the high temperatures of the denaturation step. Before the use of Taq polymerase, DNA polymerase had to be manually added every cycle, which was a tedious and costly process. Applications of the technique include DNA cloning for sequencing, gene cloning and manipulation, gene mutagenesis; construction of DNA-based phylogenies, or functional analysis of genes; diagnosis and monitoring of genetic disorders; amplification of ancient DNA; analysis of genetic fingerprints for DNA profiling (for example, in forensic science and parentage testing); and detection of pathogens in nucleic acid tests for the diagnosis of infectious diseases.
=== Modern cereal === Over 2016 to 2017, Americans purchased 3.1 billion boxes of cereal, mostly as ready to eat cold cereal. In a $9.8 billion cereal market, cold cereal purchases were 88% of the total (12% for hot cereals), with the overall cereal market declining due to reduced consumption of sugar and dairy products. Kellogg's and General Mills each had 30% of the market share for cold cereals. Honey Nut Cheerios was the leading cold cereal.

=== Separation methods === Few analytical detectors are truly specific for a single species; therefore some type of separation step is often necessary prior to detection. Moreover, separation allows for detection of multiple analytes within a single platform. Separations based upon planar chromatography (TLC) are perhaps the easiest to implement, since many μPADs are constructed with chromatographic paper. Typically, the separation channel is defined by wax-printing two hydrophobic barriers. Electrochemical detection is perhaps most common, likely due to its ease of implementation, although colorimetry, chemiluminscence, and mass spectral detection have also been used in conjunction with paper-based chromatographic separations. Despite the ease of implementation, planar chromatography is hindered by relatively low plate height (i.e., poor separation efficiency). Since the Chakraborty group demonstrated the feasibility of electrokinetic flow on μPADs, several applications of electrophoretic separations on μPADs have appeared in the literature. The Crooks group at UT-Austin successfully demonstrated that electrophoretic separations on μPADs could be accomplished at relatively low applied voltages compared to conventional electrophoretic devices due to the high field strengths that can be generated on very thin (180 μm) sheets of origami paper. Simpler separation methods can also be used on μPADs, for instance, the Henry group demonstrated the separation of plasma from whole blood using blood separation membranes.

It was among the first such works to be published, and was followed by many books by white veterans of the war. The book has been widely cited, with historians and commentators arguing that it clearly illustrated the tactics used by the Rhodesians. It does not mention any of the atrocities committed by the Selous Scouts. Many other books have since been published about the Selous Scouts. These works often glorify the unit. In 2018 The New York Times reported that glorification of the Selous Scouts formed part of online nostalgia for Rhodesia and had been taken up by far-right movements that were sympathetic to the white Rhodesian regime. Items branded with the slogan and insignia of the Selous Scouts were available for sale, and a company called the "Selous Armory" was selling a range of apparel glorifying the Rhodesian military. In 2021, the newly established Ranger Regiment in the British Army adopted a cap badge that was similar in appearance to the Selous Scouts' and may have been based on it. The Telegraph reported that "numerous officers have raised concerns" over the badge. The British Army stated that the badge was "designed around the peregrine falcon" and not the osprey as used by the Rhodesian badge.
are neutrally buoyant or nearly so; in fact most cephalopods are about 2–3% denser than seawater), they achieve this in different ways. Some, such as Nautilus, allow gas to diffuse into the gap between the mantle and the shell; others allow purer water to ooze from their kidneys, forcing out denser salt water from the body cavity; others, like some fish, accumulate oils in the liver; and some octopuses have a gelatinous body with lighter chloride ions replacing sulfate in the body chemistry. Squids are the primary sufferers of negative buoyancy in cephalopods. The negative buoyancy means that some squids, especially those whose habitat depths are rather shallow, have to actively regulate their vertical positions. This means that they must expend energy, often through jetting or undulations, in order to maintain the same depth. As such, the cost of transport of many squids are quite high. That being said, squid and other cephalopod that dwell in deep waters tend to be more neutrally buoyant which removes the need to regulate depth and increases their locomotory efficiency. The Macrotritopus defilippi, or the sand-dwelling octopus, was seen mimicking both the coloration and the swimming movements of the sand-dwelling flounder Bothus lunatus to avoid predators. The octopuses were able to flatten their bodies and put their arms back to appear the same as the flounders as well as move with the same speed and movements. Females of two species, Ocythoe tuberculata and Haliphron atlanticus, have evolved a true swim bladder.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Major regulatory agencies have not approved it for any indication. Some countries permit it only under prescription frameworks, while others classify it as a controlled substance.
