=== Earliest forms of cooling === The seasonal harvesting of snow and ice is an ancient practice estimated to have begun earlier than 1000 BC. A Chinese collection of lyrics from this time period known as the Shijing, describes religious ceremonies for filling and emptying ice cellars. However, little is known about the construction of these ice cellars or the purpose of the ice. Tang dynasty (618 AD) used saltpetre scraped from walls to produce ice in summer. The next ancient society to record the harvesting of ice may have been the Jews in the book of Proverbs, which reads, "As the cold of snow in the time of harvest, so is a faithful messenger to them who sent him." Historians have interpreted this to mean that the Jews used ice to cool beverages rather than to preserve food. Other ancient cultures such as the Greeks and the Romans dug large snow pits insulated with grass, chaff, or branches of trees as cold storage. Like the Jews, the Greeks and Romans did not use ice and snow to preserve food, but primarily as a means to cool beverages. === Aldoses and ketoses === Monosaccharides which contain an aldehyde group are known as aldoses, and those with a ketone group are known as ketoses. The aldehyde can be oxidized via a redox reaction in which another compound is reduced. Thus, aldoses are reducing sugars. Sugars with ketone groups in their open chain form are capable of isomerizing via a series of tautomeric shifts to produce an aldehyde group in solution. Therefore, ketones like fructose are considered reducing sugars but it is the isomer containing an aldehyde group which is reducing since ketones cannot be oxidized without decomposition of the sugar backbone. They are described at being cerebriform in shape, and can be diploid or tetraploid. It also contains a large nucleus with a minimum cytoplasm. Lutzner cells are more predominant in Mycosis Fungoides, but are also found in Sézary Syndrome.
=== Active site === Eflornithine's suicide inhibition of ODC physically blocks the natural substrate ornithine from accessing the active site of the enzyme (Figure 3). There are two distinct active sites formed by the homodimerization of ornithine decarboxylase. The size of the opening to the active site is approximately 13.6 Å. When these openings to the active site are blocked, there are no other ways through which ornithine can enter the active site. During the intermediate stage of eflornithine with PLP, its position near Cys-360 allows an interaction to occur. As the phosphate of PLP is stabilized by Arg 277 and a Gly-rich loop (235–237), the difluoromethyl group of eflornithine is able to interact and remain fixated to both Cys-360 and PLP prior to transimination. As shown in the figure, the pyrroline ring interferes with ornithine's entry (Figure 4). Eflornithine will remain permanently bound in this position to Cys-360. As ODC has two active sites, two eflornithine molecules are required to completely inhibit ODC from ornithine decarboxylation.
